朱一貴於康熙五十二年(1713年)隨移民潮移民臺灣,年輕時曾任衙役,後為傭工、種田為生,在鴨母寮(今高雄縣內門鄉光興村)以養鴨為業,為人豪爽好客,有“小孟嘗”、“鴨母王”之稱。
康熙六十年(1721年),鳳山縣知縣出缺,暫由知府王珍兼理,王珍把知縣職務交由他兒子管理,但王珍的兒子利用職權橫徵暴斂,引起普遍不滿。衆人遂推朱一貴為盟主,發動革命,下淡水溪檳榔林的客傢人杜君英響應革命,合作攻取臺灣府城(今臺南市);響應者有許多是明鄭時期將校武官,攻下臺灣府城後,朱一貴承襲明朝的制度,國號大明,年號永和,且廢除滿族薙發令,將旗裝脫下、長辮剪斷,恢復明朝時的服裝及傳統漢人的發式。
朱一貴占領府城後旋即稱王,名曰中興王(民間俗稱鴨母王),但後來因閩粵內哄分裂,導致其近二個月即遭清軍撲滅,清軍亦藉此事徹底掃蕩拔除南明殘餘軍隊;由此可知,當時即便沒有朱一貴在臺灣反清,也會有其他人反清:因為朱一貴事件接近可識為是清朝暗中慫恿製造,朱一貴是不自知被利用當“引蛇(南明殘軍)出洞”工具。
朱一貴雖為草莽英雄,但識見、氣宇亦有過人之處,被捕時面對平亂之福建水師提督施世驃(收臺居鹿港施琅之子,施公案施世綸之弟),猶昂然自立,自稱孤傢,後被解送北京凌遲處死。
民間傳說朱一貴死後被玉皇大帝封為“臺南小南城隍”,在臺南市小南門城隍廟被祀為主神。據說朱一貴養鴨於鴨母寮,於今高雄縣內門鄉光興村內。鴨母寮興安宮供奉朱一貴神位,興安宮在廟前水池中增置朱一貴趕鴨子的塑像。
